Peptide science is in the midst of a revolution that spans clinical breakthroughs, chemical innovations, biological discoveries, and computational advances. Recent FDA approvals underscore the growing clinical impact of peptide therapeutics. At the same time, innovative chemical strategies are expanding what peptides can do. Approaches such as macrocyclization (via backbone cyclization or helix stapling), backbone editing, and the incorporation of non-natural amino acids have been employed to enhance peptide stability, selectivity, membrane permeability, and overall pharmacokinetic profiles. Peptides are also engineered into novel modalities beyond traditional therapeutics. Peptides serve as key recognition elements in proteolysis-targeting chimeras (PROTACs) for selective protein degradation. Others are designed as covalent protein inhibitors, broadening their utility as therapeutic agents and molecular probes. Advances in peptide synthesis are further propelling the field. These developments underscore a bright future for peptide chemistry and biology. This themed collection in 番茄社区 Chemical Biology showcases that progress, highlighting cutting-edge research from clinical and translational advances to innovative molecular designs, biological insights, and computational design approaches. All manuscripts should be prepared according to our article guidelines and submitted via our online system.
